The diameter of many virus particles is usually between tens to hundreds of nanometers smaller than the wavelength of visible light, and traditional optical microscopes are limited by the diffraction limit and cannot directly observe them. However, sample preparation for TEM is laborious, which limits its application in the field of virology.
