Cell viability is a measure of the proportion of live and healthy cells in a cell population. Cell viability assays are used to determine the overall health of cells, optimize culture or experimental conditions, and measure the survival rate of cells after treatment with compounds, for example during drug screening. Generally, cell viability assays provide a reading of cell health by measuring metabolic activity, ATP content, or cell proliferation. Cytotoxicity assays can also be used to assess cell viability, which provides readings of cell death markers, such as loss of membrane integrity.

Details Dead cells can nonspecifically bind to many reagents, resulting in false positive results. Therefore, removing dead cells from flow cytometry data is a key step to ensure the accuracy of results and analysis, so as to more accurately distinguish between living cells and dead cell populations. High-content imaging can provide superior single-cell analysis capabilities and lightning-fast data acquisition time. It can perform inactive analysis of intact, fixed or live cells from monolayers to spheroids. The principle of the trypan blue method is that living cells do not absorb blue dye and dead cells will absorb blue dye. Cell viability can be calculated by the ratio of total viable cells/total cells. Staining also facilitates the visualization of the overall cell morphology.
