Imaging-based Analysis of Cellular Transfection Efficiency
Transfection is a method of introducing exogenous nucleic acid into cells to produce genetically modified cells. It is used by researchers to study the function and regulation of genes. In addition, transfection can also be used to identify protein functions. Transfection efficiency is the proportion of cells that have obtained foreign elements in a sample, and it can be evaluated using a variety of different methods. Among them, automated imaging and analysis tools are effective and robust methods for measuring transfection efficiency.

Imaging-Based Analysis of Transfection Efficiency
Time-lapse imaging of live cells is a powerful method for monitoring the transfection process at any time. Compared with transfection efficiency analysis methods such as Western blotting, flow cytometry, or real-time qPCR, live cell imaging can provide a more direct way to evaluate the efficiency of cell transfection, and can quickly provide reliable results, while requiring minimal cost.
Imaging-Based Analysis of Transfection Efficiency Workflow
CD BioSciences combines bright field and fluorescence imaging to quickly and directly quantify cell viability and transfection efficiency. It is non-invasive, automated, and avoids the use of nucleic acid stains. Therefore, the imaged samples can also be used in multiple downstream applications. The following is the workflow of imaging-based analysis of transfection efficiency.
Culture the GFP-transfected target cells in a medium supplemented with 10% FBS in a humid environment at 37°C.
Inoculate transfected and untransfected cells in different mixing ratios into 96-well tissue culture plates, and then allow the cells to adhere overnight.
Treatment of cells
Samples with different transfection efficiencies were stained with a stain at room temperature for 30 minutes.
Image acquisition and analysis
Image collection is performed on the processed cells, and the collected data is analyzed with image analysis software.
